Objective:To compare the efficacy of tamoxifen (TAM) 1 mg, tamoxifen (TOM) 10 mg, and tamoxifen plus dexamethasone (TDA) for patients with breast cancer. Methods: We retrospectively analyzed the records of patients with invasive breast cancer (tumor size <20 mm) who received tamoxifen (TAM) 1 mg, tamoxifen (TOM) 10 mg, and tamoxifen plus dexamethasone (TDA) over a 10-year period. Results: In this study, a total of 792 patients who received a combined therapy with tamoxifen (TAM) 1 mg, tamoxifen (TOM) 10 mg, and tamoxifen plus dexamethasone (TDA) were enrolled in our clinic. The patients were randomly divided into two groups: group 1 (n = 562) and group 2 (n = 569). The primary end point of this study was the occurrence of an adverse event (AE), which was defined as an incidence of 1% in the group of the patients receiving tamoxifen (TAM) 1 mg, tamoxifen (TOM) 10 mg, and tamoxifen plus dexamethasone (TDA) for a 10-year period. A total of 592 patients in both groups experienced an AEs. The AEs occurred in 4.5% of patients, and the most common AEs (≥1%) were headache, abdominal pain, myalgia, and rash. The occurrence of AEs was significantly higher in the group receiving tamoxifen plus dexamethasone (p = 0.03) than in the group receiving tamoxifen (p = 0.003) and tamoxifen plus tamoxifen (p = 0.001). Conclusion: We conclude that tamoxifen is effective in the treatment of invasive breast cancer patients with an AEs. The AEs of this study have been reported to occur at a rate of 7.5% (2/569). However, the occurrence of AEs in patients who received tamoxifen (TAM) 1 mg, tamoxifen (TOM) 10 mg, and tamoxifen plus dexamethasone (TDA) over 10 years was significantly higher than that in patients who received tamoxifen alone (p = 0.003). The occurrence of AEs was significantly higher in patients who received tamoxifen plus dexamethasone (TDA) than in those who received tamoxifen alone (p = 0.01).
